How a peptide's purity is analyzed step by step. Protocol based on USP guidelines and adapted for synthetic peptides.

Step 1

Sample preparation

Dissolution in mobile phase with 0.22μm filtration to remove particles that interfere with the chromatographic reading.

Step 2

Chromatographic separation

Reversed-phase C18 column (250×4.6mm, 5μm). Acetonitrile/water gradient with 0.1% TFA. Flow 1.0 mL/min.

Step 3

UV detection at 220nm

Wavelength optimized for peptide bonds. 20μL injection with a diode-array detector (DAD).

Step 4

Purity calculation

Area-normalization method: % purity = (main peak area / total area) × 100. All impurities >0.1% are reported.
